Ornithine decarboxylase (ODC) is a pyridoxal 5$\sp\prime$-phosphate dependent enzyme. Its product, putrescine, is a precursor to the polyamines, spermidine and spermine, which are important for cell proliferation. ODC has been purified from the slime mold Physarum polycephalum, a simple eukaryote. The holoenzyme is produced by the formation of a Schiff base between pyridoxal 5$\sp\prime$-phosphate and a unique lysyl residue of the apoenzyme. Coenzyme was covalently attached to the enzyme by reduction with KBH$\sb4$, inactivating the enzyme. Following modification of sulfhydryl groups with iodoacetdmide and digestion with trypsin, peptides were separated by reverse phase high performance liquid chromatography under acidic conditions. The pyridoxyllysyl peptide was detected by its characteristic absorbance. Because the spectrum of the pyridoxyl moiety is pH dependent, it was found to be critical to use the pH-appropriate wavelength when limited material is available. As demonstrated in this work, sensitivity is improved ten-fold by use of 300 nm for detection of pyridoxyllysyl peptides at pH = 1.5-2; many recent similar studies have continued to use absorbance at 325 nm, a wavelength appropriate for detection at neutral pH. Following purification, amino acid analysis of the peptide and synthesized pyridoxyllysine showed that the peptide does contain a pyridoxyllysyl residue. Mass spectrometry showed that the molecular weight of the peptide is 2090. The N-terminal amino acid sequence of the peptide was found to be NALPDTQPFYAVXCNPGK, where X represents the pyridoxyllysyl residue. This is the first example of an amino acid sequence from the pyridoxal 5$\sp\prime$-phosphate binding site of eukaryotic ODC obtained using absorbance to detect the cofactor directly. The determined sequence is homologous to ODC sequences from other species, including the pyridoxyllysyl peptide from mouse (detected by a less direct method) and other sequences only predicted from cDNA.
