Maize seedlings up-regulate heat shock protein genes including the small heat shock protein genes, and down-regulate other genes in response to environmental changes. DIG-labeled antisense RNA in situ hybridization revealed that hsp18 mRNA was localized to younger meristematic cells of the root-tip and shoot-tip and also to densely cytoplasmic cells of the vasculature. Radicles shifted from preshift temperatures to several shifted temperatures, produced hybridization signals only above 35$\sp\circ$C. Cells near the cap initial region, immature stele and proximal area of vascular differentiation were observed to have a high hybridization signal. mRNA induction was found also in radicles treated with zinc and cadmium, localized to the epidermal regions. The data suggest that 35 $\pm$ 1$\sp\circ$C is the thermal shift threshold temperature for the induction of hsp18 mRNA in maize. While meristematic cells respond to heat-shock, epidermal cells respond to metal-ion insults.
