The aim of this study was to engineer a novel biomaterial capable of supporting vascularization to repair injured cardiovascular tissues. The limited transport capacity of oxygen and nutrients into thick tissues can be overcome by inducing vascularization within engineered tissues via angiogenesis in vitro, thus improving the survival of tissues. To promote angiogenesis, vascular endothelial growth factor (VEGF) and angiopoietin-1 (Ang1) were covalently immobilized onto collagen scaffolds using 1-ethyl-3-[3-dimethylaminopropyl]carbodiimide hydrochloride (EDC) chemistry. Over 3 days and 7 days in vitro, seeded endothelial cells had increased proliferation and metabolism on scaffolds with immobilized growth factors compared to PBS and soluble controls. Hematoxylin and eosin, live/dead, CD31, and von Willebrand factor staining all showed improved tube formation by endothelial cells with co-immobilized VEGF and Ang1. Immobilization of VEGF and/or Ang1 also increased blood vessel density in the chick chorioallantoic membrane assay and the rat right ventricular outflow tract model in vivo.
