Abstract: Microbially secreted GHs are responsible for the degradation of various polysaccharides across environments. The organization of GHs and other carbohydrate-active enzymes into the CAZY-database (CAZY) has guided interest into the field of CAZYmes by providing functional descriptions of various CAZYmes, however CAZY provides no tool for sequence comparison or sequence download. Here, we extracted the characterized sequences listed on CAZY via the listed NCBI-portals, and analyzed the sequences using the GeneHunt approach. First, we validated GeneHunt on all of the biochemically-characterized sequences listed on CAZY, where GeneHunt successfully (re)identified over 96% of biochemically characterized GH sequences. Next, we used GeneHunt on the entire M5nr database, where most GH sequences were detected as SDGH with few MDGH and MAGH. Then subsequently, we used GeneHunt to search for GHs in 129 assembled metagenomes, where we were able to demonstrate the activity of a GH5-GH10 (cellulase-xylanase) MAGH.
